Jeho prarodiče byli varietní umělci a otec pracoval v reklamním průmyslu. Když mu bylo šestnáct, v londýndýnském divadle Old Vic zhlédl inscenaci Macbetha a v tu chvíli se rozhodl, že se stane hercem. Poté studoval dva roky v Londýně na Royal Academy of Dramatic Arts a hrál v divadelním souboru National Youth Theatre, se kterým hostoval až v Austrálii.

Ve svém angažmá v Birmingham Repertory Theatre na sebe upozornil mimojiné ve hře G. B. Shawa Svatá Jana. Postupně se představil na různých londýnských scénách a později, jako člen Royal Shakespeare Company (Královská Shakespearovská Společnost) slavil úspěch v Shakespearově dramatu Romeo a Julie. Za zapsání do širšího diváckého povědomí vděčí především TV seriálu Saturday While Sunday(1966), díky němuž dostal roli ve svém prvním celovečerním filmu. Až rok 1987 s sebou přinesl značný vzestup jeho popularitym, když ho producent Cubby Broccoli obsadil do role čtvrtého (Sean Connery, George Lazenby a Roger Moore) představitele úlohy pověstného tajného agenta 007 Jamese Bonda ve snímku James Bond - Dech života (1987, režie John Glen). Beginning his career on stage, he made his film debut as Philip II of France in the 1968 historical drama The Lion in Winter. He took roles in the period films Wuthering Heights (1970), Cromwell (1970), and Mary, Queen of Scots (1971). Dalton also appeared in the films Flash Gordon (1980), The Rocketeer (1991), Looney Tunes: Back in Action (2003), Hot Fuzz (2007) and The Tourist (2010).

On television, Dalton played Mr. Rochester in the BBC serial Jane Eyre (1983), Rhett Butler in the CBS miniseries Scarlett (1994), Rassilon in the BBC One sci-fi series Doctor Who (2009–2010), Sir Malcolm Murray on the Showtime horror drama Penny Dreadful (2014–2016), and the Chief on the DC Universe / HBO Max superhero series Doom Patrol (2019–2021). He portrayed Peter Townsend in the fifth season of The Crown.

C'est en jouant la pièce MacBeth à l'âge de seize ans que Timothy Dalton se découvre une vocation de comédien. Dès lors, il étudie le théâtre au Royal Academy of Dramatic Art et rejoint ensuite le Birmingham Repertory Theater. En 1966, il fait sa première apparition télévisée dans la série Sat'day while sunday, où il côtoie le jeune Malcolm McDowell, et deux ans plus tard il se retrouve à donner la réplique à Peter O'Toole dans le médiéval Lion en hiver (1968).

Jugé trop jeune pour incarner James Bond dans Au service secret de sa Majesté (id.), il continue dans le film à costumes en jouant le prince Rupert dans Cromwell (1970) de Ken Hughes et le Lord Henry Darnley dans Marie Stuart (1971) de Charles Jarrott. Tout en travaillant l'œuvre théâtrale de William Shakespeare, Timothy Dalton se lance dans une carrière internationale, apparaissant dans El Hombre que supo amar (1978), drame historique espagnol se déroulant au temps de l'Inquisition, et dans la super production américaine Flash Gordon (1980).

Après avoir interprété Rhett Butler dans la série télévisée Scarlett (1994) et l'empereur Jules César dans le téléfilm Cléopâtre (1999), Timothy Dalton fait son retour au cinéma en prenant les traits d'un shérif local aux prises avec des gangsters dans le polar Fausse donne (id.).

Quatre ans plus tard, il s'auto-parodie en agent secret, père de Brendan Fraser, dans Les Looney Tunes passent à l'action (2003), une comédie d'aventure de Joe Dante mêlant acteurs réels et personnages de dessin animé. Seine Bond-Interpretation wurde von vielen Kritikern und Lesern der James-Bond-Romane von Ian Fleming als dem Charakter der Buchvorlagen am nächsten eingestuft. Zum Zeitpunkt seines Engagements waren auch bereits Pierce Brosnan, der vertraglich anderweitig gebunden war und später Daltons Nachfolger wurde, sowie Lewis Collins für die Rolle im Gespräch. Entgegen unterschiedlichen Berichten über Streitereien mit dem Regisseur oder eine Kündigung Daltons gab die Produktionsfirma bekannt, dass es Daltons Entscheidung war, seine Rolle als Bond aufzugeben. Gerne hätte sie es gesehen, wenn er seine Rolle fortgesetzt hätte.

In der Fortsetzung Scarlett des Klassikers Vom Winde verweht als Mini-Fernsehserie übernahm Timothy Dalton im Jahre 1994 die Rolle des Rhett Butler, die vormals von Clark Gable gespielt worden war. Seitdem trat er auch verschiedentlich in weiteren Fernsehserien wie etwa 1999 in Cleopatra als Julius Cäsar auf. Einem jüngeren Publikum dürfte Dalton durch seine Darstellung des mordenden Supermarkt-Leiters Simon Skinner in der britischen Komödie Hot Fuzz – Zwei abgewichste Profis oder durch die Rolle des strengen Polizisten Jones in The Tourist bekannt sein.
