Het is het jaar 2021. Diep onder het oceaanoppervlak doemt een enorm, magnifiek hightech complex op: Sealab. Een multinationaal wetenschappelijk station met een jaarlijks budget van in de biljoenen, bemand door een bonte verzameling ontevredenen en sukkels die ongeschikt waren voor werk in de particuliere sector. Ze laten eigenlijk geen onderzoek doen, maar brengen hun tijd door met onderling kibbelen of gewoon gek doen. De bemanning heeft hun ongelukkige leider, kapitein Murphy, tot onderwerping gemanipuleerd en is tevreden met het besturen van de overheidsklok, waarbij ze dikke, gevaarlijke loonstrookjes binnenhalen.

The year is 2021. Deep below the ocean's surface, looms a vast, magnificently high-tech compound: Sealab. A multi-national scientific station with an annual budget in the trillions, manned by a motley collection of malcontents and screw-ups who were unfit for work in the private sector. They really don't get any research done, but instead spend their time bickering among themselves or just plain goofing off. The crew have manipulated their luckless leader, Captain Murphy, into submission, and are content to ride the government clock, raking in fat, hazardous-duty paychecks.

A műsor a 70-es években bemutatott Sealab 2020 gyerekműsor paródiája. Az eredeti műsorhoz hasonlóan a Sealab 2021 is egy vízalatt dolgozó csapat kalandjait követi nyomon a tenger felfedezése közben, azonban sokkal több felnőtteknek szánt jelenettel és humorral megtoldva azt. A műsorban többször is utalnak más Adult Swim sorozatra, többek közt a Space Ghost Coast to Coast-ra is. A sorozat nemcsak a Sealab 2020-at parodizálja, hanem az 1970-es évek gyerekeknek szóló rajzfilmsorozatait is összességében.

No Sealab 2020, a tripulação do laboratório submarino pesquisava sobre a vida marinha e combatia eventuais perigos, como poluição e caçadores. De acordo com Sealab 2021, a tripulação ficou louca após permanecer um ano presa dentro do laboratório, e pouco a pouco foi deixando de lado as pesquisas e o trabalho em geral e começaram a gastar a verba com futilidades, entre outras atitudes estúpidas. Assim como grande parte das animações do Adult Swim, a série usa o nonsense, o que leva a, entre outras coisas, ignorar a continuidade, já que o laboratório explode no final de quase todos os episódios.

A tripulação da clássica série Laboratório Submarino 2020 foi substituída por uns inúteis que, por alguma razão, se autodenominam cientistas.

Действие сериала происходит через год после событий Морлаб 2020. За время этого года экипаж станции постепенно сошёл с ума и теперь проводит всё своё время в безделье и безумных развлечениях, а не в исследовательской работе. Сериал не придерживается связей между сериями, и почти все серии представляют из себя независимые истории, сама станция в конце многих серий уничтожается, а персонажи умирают самыми ужасными и мучительными путями.
